A postdoctoral position is available in Ralph Scully’s lab at the Beth Israel Deaconess Medical Center and Harvard Medical School to study relationships between DNA repair, genomic instability and cancer in mammalian cells. We use a variety of genetic, biochemical and cell biological tools to analyze these relationships. A key novelty in our research toolkit is the use of site-specific replication fork barriers, which enable us to study ‘replication stress’ and its consequences at a single, defined locus of the mammalian genome. This approach has yielded a model system for studying the Fanconi anemia pathway of stalled fork repair in mammalian cells (Nature 2014, PMID: 24776801). It also led to our discovery of the mechanism underlying specific genomic instability patterns in BRCA1-linked cancer (Nature 2017, PMID: 29168504). Analysis of genes that cooperate with BRCA1 to suppress genomic instability led us to identify a synthetic lethal interaction between mutations in BRCA1 and FANCM (Molecular Cell 2021, PMID: 33882298). Our unpublished work suggests that FANCM is a promising therapeutic target in BRCA1-linked cancer.
The new postdoc position entails exploring new approaches to the genetic regulation of stalled fork repair, and the development of new methods to study stalled fork remodeling. Some previous experience in DNA repair/genomic instability is desirable but not essential. Core competency in basic molecular biology techniques is essential, as is an enthusiastic commitment to engaging in cutting-edge research.
