An NIH funded post-doctoral position is available in the laboratory of Prof. Maria Pellegrini in the Chemistry Department at Dartmouth College. Research in the group aims to understand and modulate protein-protein interactions within the NF-kB pathway to develop new therapies for cancer, inflammatory and autoimmune diseases. We are interested in the IKK complex, a central node in the pathway and in the interactions between the scaffolding protein NEMO and its partners, particularly the kinase IKKbeta. NEMO is a long, predominantly coiled-coil protein which is refractory to structure determination, and our lab has engineered constructs suitable for both X-ray crystallography and NMR spectroscopy. Structures are determined by X-ray crystallography, SAXS and NMR spectroscopy (see: https://pubmed.ncbi.nlm.nih.gov/31929506; https://pubmed.ncbi.nlm.nih.gov/30814588; https://pubmed.ncbi.nlm.nih.gov/25286246). The structures constitute the basis for protein-protein inhibitor design, which relies on computational screening, NMR-based screening and structure-based optimization. The research develops through expression and purification of proteins and complexes, biophysical and biochemical characterization through CD, AUC and DLS and binding assays (pull-downs/Co-IPs, ELISA, FP, SPR, ITC).
Applicants should have a recent Ph.D. and solid training in biochemistry and protein chemistry. Previous research experience in structural biology is preferred.
Our lab is located in an open space shared with two other structural biology research groups within the Chemistry Department, fostering collaboration and a highly cooperative community. Our group has access to first rate facilities within the department and at the Institute for Biomolecular Targeting (@DartBioMT), including protein production and characterization, imaging, microscopy and the high-throughput crystallography core facility. We manage and extensively utilize the Biomolecular NMR Facility at Dartmouth, equipped with 600 and 700MHz instruments, cryoprobes and SampleJets (https://sites.dartmouth.edu/mierkelab/nmr-facility/). We have regular beam time at the NSLSII FMX, AMX and LiX beamlines. We are also part of the Cancer Biology and Therapeutics research program of the Norris Cancer Center, an NCI-designated Comprehensive Cancer Center (@CancerDartmouth).
